Dear Histology Experts:
We are about to embark on a huge project and wish to save time by using wax pens to provide an enclosed area in which to place our reagents during staining (previously we carefully dried around our sections to provide a way of creating a meniscus by which the surface tension retained the reagent on the tissue at the desired concentration).
But we now need to speed up!!! However when we have tried wax pens in our protocols this has left autofluorescent regions on the slide which impact our automated imaging platforms ability to set exposure times.
Any experience or advice would be gratefully received.
