Dear all,

I´m planing to transduce white pre-adipocytes with a lentiviral constructs for inducible shRNA-mediated knockdown of certain target genes. Since my target genes are strongly involved in lipid synthesis and thus adipocyte differentiation, I would need an inducible system that has a tight promotor to avoid differentiation defects due to "background" target shRNA expression. At the same time the system should ideally also be sensitive to the inducer to have efficient knockdown of the respective target genes after adipocyte differentiation.

Has someone experience with the IPTG- or Cuamte-inducible systems? I would like to avoid Doxycycline, because it is suspected to have an impact on neutral lipid metablism.

I would really apprechiate to know your opinions and experiences with these systems.
