I am trying look for progesterone receptor A and B levels across gestation in uterine nuclear extracts by (NuPage) western blotting. I tried antibodies from different companies but nothing worked positively. Does total homogenate give good signals for PR A/B compared working with nuclear extract? What changes do I need to make in the protocol if I use RIPA buffer for extraction?

I tried antibody from Santa Cruz and Cell signalling but did not work, now trying out one from Pierce MA1-410. Any tips to get good clear bands for PR A and B ? whether whole cell lysate is better than nuclear protein extract to test these antibody. Any idea why these antibodies are not working?
