Actually i have not faced this type of problem. but in my opinion you have increase the amount of B-mecaptoethanol and the Chloroform:isoamyl alcohol thrice to remove polyphenols. try this may be helpful. gud luck
If you have sodium metabisulfite in your lab, that can do the trick for you. Its more potent than PVP in blocking polyphenolic compounds. hope this helps
I really had problems with extracting RNA from elm trees.. My problem was polysaccharides but this protocol really helped.. Also applies for polyphenols it uses boric acid in the homogenization/extraction buffer.. Maybe you should also use Alcohol Precipitation not Isopropanol if you want DNA. In my samples DNA precipitated right after adding of Isopropanol....
