Hi, there are many protocols in different papers but it is hard to know which one would be reliable. I am looking for measuring Total phenol content, Total Flavonoid Content and Antioxidant activity in ''ETHANOLIC extract'' of leaves and flowers of a plant. Many thanks in advance.
I think you can employ the method as described by Singleton el al. (1999) as seen in the attached publication.

You might able to get all the protocol which is very easy to do.
Please don't bother about the reliability of the protocol, do whatever suitable for your plant sample. As you are working with Elaeagnacea family plants, it will be better if you search some review paper related to the composition of the phytochemicals present in the Elaeagnacea family plants. As well as its medicinal use.
It will help you to select the suitable protocol for your experiments.

My dear freind, you can not be so confused with the different protocols. Read and find out what was the purpose of that investigator in using a particular protocol. After that see whether your requirements is similar to that person's and try it out. As a third person, it is rather in appropriate to advice you on such a subject.
