How can I label the mouse cells? I have some mice with human tumor xenograft, and need to distinguish the human tumor cells and mouse cells by flow cytometry. I can label the human cells by HLA antibody, but is there anyone that knows what kind of antibodies I can use to labeled the mouse cells? I mean all the mouse cells, including epithelial cells, endothelial cells, immune cells.... but not a specific group of cells.
Someone suggested H2Kd to me, but athymic nude mice are not an inbred strain or BALB/c source, so it doesn't work in the pilot study.

If your tumor cells have HLA antigens, you can use just anti- human HLA A,B,C antibody for detection of tumor cells. You don't needs the others. Because, mouse cells don't express human HLA antigens. If tumor cells no have any MHC molecules you can not use this method.

what about a mouse beta2microglobulin antibody? This one should recognize all MHC I molecules (in contrast to H2Kd which only works in certain strains of mice).

Thank you very much for your advice. I'm considering the mouse cell marker because of two reasons. Firstly, I'm testing the samples from mouse xenograft, and there're a lot of non-cell particles in the sample, such as fibers, debris..., I want to see if I can exclude such junks by both human cell and mouse cell staining. Also, I'm doing studies on nanoparticles, and I want to calculate the total particles in total mouse cells. Do you think I should do the mouse cell staining for such purpose?

sorry for my late reply. I was in outside of my country . I didn't see your message. If you use Balb/c mice, I suggest H-2Kd or H-2Dd specific antibody for discrimination of mice cells. If your nanos have any fluorescence, your should choose a different fluorochrome for can detect nanos and mice cells together.
