I am looking for an anti-P-src antibody which interact with human CD44. Tyr 418 was used in the literature but only Tyr 416 on Cellsignaling website. Are they the same?
I got something from the product information of Anti-phospho-SRC (pTyr418) {cat#S1940, Sigma }.
"Autophosphorylation of chicken c-Src also occurs on Tyr416 (Tyr414 on frog c-Src, Tyr418 on human c-Src, and Tyr423 on mouse c-Src). and is readily demonstrated in vitro. Phosphorylation of Tyr416 is mostly intermolecular since its rate is concentration-dependent. This site is located in the catalytic domain of human Src,6 and phosphorylation of this site is required for full catalytic activity"
But I am still confusing, for example, anti P-src tyr416 also reactive with human, monkey, rat and mouse. ( cell signaling #6943).
For my experiment, I plan to see the activation of src in human epithelial cells line. May I use Anti-phospho-SRC (pTyr416) which I already have one vial or I need to buy a new Anti-phospho-SRC (pTyr418)?
In Src sequence, there is only one major autophorylation site, Y416, (which could also been 418, 419, 425, depending on isoform and species) reflecting the activity of the kinase.
Src sequence is well very conserved so if your antibody reacts with Human Src, it should work!!
