I'm trying to optimize process of DNA extraction from dried blood spots on FTA cards for qPCR or PCR, for diagnostic purposes.

I've tried Qiagen DNA mini kit, and Alkaline extraction. I've also tried pre-soaking the cards overnight in PBS before extraction. I know that you can wash cards and put them directly in PCR mix but I am hoping to get qPCR data.

The average yield I get using Qubit, is 0.1-0.8ng/ul in 100ul elution volume. This may be enough for qPCR, but I just don't think with so little DNA that the diagnostic would be that sensitive.
