I am just curious to know what will happen if Tcells and cancer cells that are not HLA matched are co-culture together? Will the Cancer cells die due to allo reaction (GVHD) or will there be no effect at all?
Hi Sasidharan! As Tomas Koltai said, it is difficult to give an answer just in theory. Basically, tumor cell killing by T cells depends on the formation of the immune synapses between T cells and target, which in turns depends on HLA expression. So, in theory, if they are mismatched, killing should not occur. However, there can be a certain GVHD, as you mentioned, but it could be also inhibited by inhibitory ligands expressed on tumor cells. Overall, it is more likely that you don't see killing, but it should be tested
