i want to deactivate almost all the virulence genes of Streptococcus agalactiae so that no transcription of those genes could happen normally. For that I am looking for simpler methods of causing mutations in the genes. kindly suggest methods ( site directed mutagenesis ) that could be used to mutate the genes through certain chemicals (mutagens) which won't require replacement of DNA fragment with any other foreign DNA fragment.

I suggest using signature-tagged mutagenesis(STM) to identify them you can use site-directed mutagenesis as you said.

Walid Hassene Hamri i read about STM and it says the approach is used to identify virulence genes, whereas, the virulence genes of streptococcus agalactiae are already known. Now I want to completely deactivate all the possible virulent genes in my sample of S. agalactiae. I have read methods of site directed mutagenesis and all of those involve great deal of genetic engineering such as suicidal plasmids vectors etc but I am looking for simpler methods which only disrupt the virulent genes in any way like deleting a piece of DNA fragment from the genes, point mutation, by using mutagens.
