Actually i am working on possibilities of horizontal gene transfer of resistant gene in broth coculture of two different types of bacteria, and the protocol i follow involves the use of MOPS buffer while exposing the culture to antibacterial agents. i want to know whether it is necessary to use MOPS Buffer for this experiment or it will make no difference? and does pH change occur during the HGT?
The protocol i follow is actually used with yeast rather than with bacteria.

I see no special reason why MOPS should be required in your case, particularly if the protocol you refer to was designed for yeast. In first approximation, I would choose a medium in which both strains between which HGT is supposed to happen grow happily. Further considerations may apply if for example some conditions favour the presence of congugation pili.
