I want to dissolve L-DOPA to treat my melanoma cells. I tried with DMSO, PBS and ethanol, but L-DOPA did not dissolve in those solvents. Then I tried with nuclease free water which dissolve L-DOPA partially, however, when I heated the solution at around 75 degree celsius for 5-10 minutes, L-DOPA was dissolved completely. L-DOPA has melting point of 284-286 degree celsius. I was a little puzzled thinking whether increasing the temperature of the solution to 75 degree celsius would degrade this drug or not!
Try saline solution, L dopa should be soluble in PBS. Its should also be soluble in water (not ethanol) without having to increase the temperature to 75 degrees. Maybe the concentration of the solution should be reconsidered?
I have had good results in coating membranes with L-DOPA dissolved in a Tris-buffer with a pH of 8.5 following a protocol developed by my colleagues. For more details, you could look into their paper below. Good luck.
