I joined a lab recently and the work here is mostly western blotting. My lab mates incubate primary antibodies for 2-3 days at 4 degrees Celsius and secondary antibodies for 1 day (overnight) at 4 degree C before band detection.

they seem to get some bands (not all the time) but when I tried to learn the western blot protocol on the internet, i couldn't find anyone that incubates antibodies for this long. i read that usually it's 1 day for primary and few hours for secondary.

should i just follow their protocol or will following the internet's protocol (shorter incubation time) get me better bands?
