The use of HPLC might give some unknown peaks. However, LCMS would allow a more detailed identification of the compounds in the hydrolysate. Such compounds might include sugars (monomers and oligomers), organic acids, ethanol, etc? I was told that C18 column is not suitable. Can anyone suggest a suitable column that would allow for the identification of the mentioned compounds in the hydrolysate?
I'm afraid it's impossible to solve your problem using one column and even one method. You can separate carbohydrates using the amino-phase and MS detection. We use for that purpose ligand exchange columns for mono- and oligosaccharides determination with RI detector as proposed Mr. Li, it is good developed and robust method. Separation of phenolics in LC usually demands C18 or bifunctional reversed stationary phases and UV or MS detection. Volatile compounds are better analysed with GC or GC-MS techniques.
Agreed with Dmitry. Because of the complexity of lingnocellulosic compounds, it is very difficult to analyse all the stuff that comes out after reaction. This requires a combination of detection systems. Volatiles are often done with GC-MS for identification, but for the heavier substances like oils and waxes, you may need to consider on-column setups.
