The IUPAC procedure for determining total cellulase activity using filter paper states that one should prepare two dilutions of the enzyme such that one dilution will release > 2 mg glucose from filter paper and the other dilution will release < 2.0 mg glucose. I have read many papers based on this method but I cannot find any that clearly explains how to go about this. How can one know the dilution of the cell-free extract that would release those amounts of glucose? Besides, the method is too complicated. Can anyone help with an explanation?
