In running an experiment to plot growth curve of an organism which is growing on a cellulosic substrate, it is necessary to collect samples from the shake flask at intervals (2-4 hours over 72 hours). Growth of the organism is to be measured from the samples by measuring OD and taking plate counts on agar plate. Cellulase activity of the samples will also be measured at the different time intervals.
Assuming I am to collect 5 ml aliquots from the shake flask at each sampling period, can I just keep the samples at -20 deg Celsius so that I can perform all the cfu, OD and enzyme activity measurements after the last sampling? Or is it better to perform the tests immediately after each sampling [which would be more stressful :) ] ?
