I am having a problem with identification of DU 145 cells. There are 3 types of DU cells in our lab. 2 are quiet similar but the third one is v. different. I am uploading the pics of all three 10X and 20 x. So, can somebody please tell me which of these is DU145? I matched these cells with ATCC but I am unable to decide. Please help.
You can never tell by just seeing the morphology of a cell, even then DU145 cells normally grow as mat (margins of cells are not distinct when it is in colonies). Although STR is the best suggestion, but it is an expensive technique costing USD 295 for verified STR testing where ATCC matches the submitted cell lines with the cell lines present in its database. And accorrding to the figures above, you have to get STR done for 6 cell lines for the confirmation of DU145.
What we know about PCa cell lines is this and you can do the following before sending the lines to ATCC, this will save your money and time:
1. DU145 and PC3 cells are AR negative while LNCaP are AR positive.
1a. So first select cell line which is AR negative, i.e. it will be either PC3 or DU145.
2. Now DU145 is ER alpha -ve, ER beta +ve, PR +ve and PS2 -ve. while PC3 is positive for ER alph, beta and PS2. DU145 also expresses higher levels of EGFR as compared to PC3.
So you will be sending the cell line for STR which is negative for AR, ER alpha, PS2 -ve and EGFR high.

As I have shown in the previous paper (Figure1 in Biochem Biophys Res Commun. 2014 Jan 3;443(1):239-45.), which focuses on EpCAM expression in PC-3 and DU-145 cells, DU-145 cells have less potential to form the spheroids as compared with PC-3 or LNCaP, other human prostate cancer cell lines.
In my opinion, the second , firth, and sixth photographs seems to resemble the morphology of DU-145 cells which I have the experience of the cell culture.
Strikingly speaking, it is important to submit RNA extract for STR confirmation.
STR; Short Tandem Repeat
Sometimes DU145 cells might seem a little different depending on the passage number and culture conditions. I suggest cell line authentication before proceeding with complex experiments.
To me slides 1 and 2 appear to look like DU-145. Also make sure your cell are clean with respect to contamination of mycoplasma or other microorganisms by using a University service that can test for contamination or purchase kits that can be used for testing.
