I am going to do ms pcr but I have doubt on water milliQ water because previously RT pcr result was not coming by using that water. So I borrowed water from another department but that water was autoclaved Depc treated milliq water. So can I use this for bisulphite conversion and ms pcr?ddepartment
For best and reproducible results you should use steril-filtered "PCR grade" water, free from nucleases or nucleid acid contaminations. Such water is available from most life science reagent vendors. Costs of about 50 EUR per liter of water appear high, but they suffice for over 100.000 PCR reactions. This water is also suitable for bisulphite conversion.
I have used sterile water which uses for injection in a single dose container and gave me a good result it is extra high quality without contamination.
Yes, it si OK, DEPC, is no longer present after sterilization. Also steril water for injection works fine.
