i wish to culture monocyte derived macrophages (MDM) from PBMC's. Poly d lysine (PDL) coated dishes are routinely used for this purpose but I would like to coat my dishes with PDL instead of buying the commercially available pre coated ones. PDL comes in different molecular weights therefore i would like to know which

1) molecular weight type 

2) Poly d lysine concentration

is ideal for culturing macrophages. 

And is it an absolute requirement for the dishes to be coated with PDL. What would happen if i culture them on FCS/FBS coated dishes or on polystyrene coated dishes which are negatively charged as opposed to the positively charged poly d lysine.?
