I want to enrich a culture medium with isolated exosomes of a given cell line for cell signaling studies in another kind of cells. What is the minimum amount of exosomes to enrich this culture medium?
I dont think there is a minimum amount. If your goal is to see an effect in the recipient cells, I recommend you to try several concentrations/amounts of exosomes. Be sure that you know how much particles or exosomal protein you have per condition. It also depends how long you preincubate your cells with the exosomes and on the uptake of your exosomes by your cells.
I agree with Iva's comments. In addition, I would like to note that the contents of the exosomes vary with culture conditions. Therefore, it is important to know what phenotype in the recipient cells would be of interest. If it is known, exosomes can be "enriched" with the mediator of this effect. For example, exosomes containing dsDNA or RNA can be prepared under a set of culture conditions.
I agree. First of all you need to do is to quantify the isolated exomes. You have to treat exosomes accordingly in trial and error method to you desired cells.
