ATCC recommends limiting up to passage 5 (cancer cell lines) for cell culture studies in order to avoid significant genetic drift from reference cell line. Every time a cell culture is subcultured the passage number increase by one. When the cell population say at passage 4 is cryopreserved, the passage no is RESET back to zero! (If this cell is thawed and cultured it is designated as passage zero). I wonder if I have got this wrong. Something doesn't seem to be right. A cell line having repeatedly cultured and cryopreserved should have genetically drifted to another continent.
Hi Abu, when you try to name your cell how many passages it's just for your convinent, If you like you can name it passage one when you thaw it from liquid nitrogen or you can follow your original passage to name it. If I got a new cell line from others I name passage one and set up my own database. Usually I follow the passage when I freeze it down before I always label how many passage on the cryo-viral. remember I always finish my experiment within 25 passages. So when you get a new cell line, always try to amplify more and freeze down more. Good luck!
