I am looking to know the IC50 of podophyllotoxin in MCF-7, HeLa or other cell lines, in literature I found as positive control etoposide or teniposide but never the podophyllotoxin.
I've worked with MCF7 cells developing some cytotoxic assays. For my experiments, I couldn't find the IC50 for the drugs that I used. So, my advice is to set up a pilot experiment, using different cell concentrations exposed at different podophyllotoxin's concentrations during 1, 3, 5, 10, 18, 24, 48 and 72 h. This can be done in a 96-well plate. You can also find a good resource how to set up these experiments in the book: "Culture of animal cells" by Ian Freshney. Hope this helps!
