Hi everyone,

I’m currently extracting total protein from mouse liver tissue using RIPA buffer supplemented with a protease inhibitor cocktail. After quantifying the protein concentration using the Bradford assay, I’m consistently getting values around 50 mg/mL, which seems quite high. We even had to dilute our samples 1:100 just to bring the absorbance within the range of the standard curve.

Is this yield typical for liver tissue, or could there be an issue with our extraction protocol or the Bradford assay itself (e.g., interference from components in RIPA)? I want to make sure we’re not overestimating due to buffer interference or pipetting inconsistencies.

Has anyone encountered similar results or have benchmarks for expected protein yields from mouse liver?Thanks in advance!
