MIC is lowest concentration at which heavy metals will inhibit the growth of the test strain. to determine MIC for heavy metals .. u ll need to add diff concentrations of heavy metals into the media and inoculate yr test strain and check for growth after overnight incubation.
MTC is the highest concentration of metal, which has no effect on the growth of the test strain. for protocol u can refer - Schmidt T, Schlegel H G, Combined

MIC is the lowest concentration of the inhibitor that produces complete absence of growth of your test organism (ex; bacteria in broth media) after sufficient incubation, it is also called lethal concentration.
Maximum tolerable concentration is the concentration at which there is no difference in the growth pattern (Ex; turbidity for bacteria) when compared to the untreated/control sample.
MIC is tested against the contaminated or undesirable species to grow but in minimum effective dose while MTC is the dose which is tolerable to the desired species and after MTC desired growing species show death pattern due to toxicity of the drugs.
MIC may be used against microbes like species to kill them in minimum dose but because every drug is toxic to the host too so MIC dose not only damage microbes but also to the host taking it.
MTC are so the maximum dose tolerable to the host.
A ideal drug should have minimum MIC and higher MTC .
Ratio of MTC wrt MIC is measurement of therapeutic index to know how high effectivenesd of a drug
@Ali you may use agar diffusion technique for MIC and tube dilution technique for MTC. The minimum inhibitory concentration that inhibit growth in agar is the MIC but maybe MTC as well. To verify, culture the test organisms in serial dilutions of the test metals based from your MIC and check for growth. The least dilution is assumed to be MIC. Streak a sample of the MIC into fresh agar, if it grow then your MIC is also your MTC. Go farther for concentrations higher than your MIC till you reach your MTC. Good luck!
