I've been trying to silence a particular gene in my rhabdomyosarcoma cell lines (Rh41, Rh30 and Rh5) with FuGene HD Transfection Reagents from Promega and siRNA from Dharmacon, but up till now I have not found the optimal condition to transfect these cell lines. Does anybody have experience with these cell lines and know what the best conditions are?

I would like to offer you the possibility to try alternatives to your transfection reagent to silence gene expression : our Lullaby siRNA transfection reagent. This reagent is totally biodegradable and really efficient.
Lullaby is dedicated to gene silencing; among numerous publications, there is one from the Cancer Research UK Beatson Institute of high importance.
The author stated :"Often we find that reagents suggested by collaborators are effective at transfecting siRNA but demonstrate associated toxicity at the concentrations we require. By far, our preferred reagent is Lullaby from OZ Biosciences. We have used this reagent in over 20 cell lines and have found it essential in enabling siRNA screens in hard to transfect cell lines such as those derived from GEMM pancreatic tumours and AML suspension cell lines, with minimal toxicity."
so I do think this should be worth trying it in your cell model.
