What is the best way to construct a microdialysis calibration curve?

Do you dilute your protein of interest in the perfusion buffer/serum/etc? Has anyone tried spiking the mouse?

Can you please give more detail? What are you doing microdialysis on? What is the protein of interest? What is your method of measuring this protein from micro dialysis experiments once you have collected whatever fluid you are collecting?
