I am looking for a vital dye - preferably quick & easy as opposed to fluorescent dyes - for sporangia viability testing for downy mildew (Peronospora farinosa).
Thank you both for the publications - I have seen these but most of them are fluorescent dyes and I am trying to avoid using those set up. Any other recommendations?
Hi - unfortunately the sporangia has to be stained in some way to differentiate between viable and dead sporangias, it is not doable only by looking at them. I am doing a contrast between MTT, Phloxine B, Trypan Blue but all of them have their drawbacks when used for sporangia (they have been more-or-less reliable for oospores). But I am looking for other stains as well (non-fluorescent) that could perhaps outperform the 3 other dyes.
