I have isolated actinobacteria from soil, but until now I only get one actinobacteria. Most of microorganism grow there is bacteria. I used Nystatin, penicillin, and sometime use benomyl. Did I use a wrong supplement?
you didn't described wich supplements you have used in the media. Any way you must use specific media for isolating actinbacteria supplemented with Cycloheximide (40micrograms per ml) to prevent the growth of fungi and nalidix acide to prevent the growth of most Gram negative bacteria (10 to 20 microgams mer mL
Just check if there are difference in the bacteria's cell membrane or what type of protection actinobacteria has. The first thing to do in research is knowledge of the subject don to molecule level and if it is organism then the metabolism.
In general, there are media for selecting actinobacteria and many commercially exist . Generally they are poor media. You can add glycerol as it is exclusively used by actinos. Olson medium is notably a good one. Media for isolating actinobacteria should contain an inhibitor of gram negative bacteria and fungal inhibitors. You should avoid inhibitors of gram positive bacteria or any with broad activity.
This is possible by using Actinomycetes isolation agar by usin glycerol as source of carbon, Nystatin to inhibit fumgi and Ciprofloxacin to inhibit bacteria.
Hai, there are many papers on selective isolation of actinomycetes. First before you decide on antimicrobial agents to be added to your media - you can consider pre-treatment such air drying, moist or dry heat treatment of your environmental samples to cut down the number of contaminating microbes on the isolation plates. Do email me at [email protected] if you need help. Am attaching one paper for you.
how is your process for the sample treatment. First, you have to dry your soil in the sun or dry at 40 -50oC because Actinobacteria can stand at a higher temperature than bacteria. Then, the soil is diluted and spread on a suitable media. However, you have to take your Actinobacteria to the new media as soon as they grow
As we are well aware that actinobacteria are similar to Gram positive eubacteria, selective agents can not solve your problem but as also suggested by V.Sabratnam the use of actinomycetes Isolation agar(Himedia) can be very helpful in isolating the organisms you are looking for.
