I have to do antibiotic protection assays with Calu-3 cells. These are growing well in cell culture flasks but not in wells of plates. Can anyone tell me the optimal seeding density for calu-3 cells in 6-well and 24-well plates?
Calu-3 cells do not grow well if plated at too low density. In general, you plate 1-2x10^5 cells in 6-well plates and 10-15x10^3 cells in 24-well plates. Split at 1:2 or 1:3, higher dilutions do not work well.
If I were you, I would try a couple of plates with different amounts of cells and see which one works best for your needs.
Thanks Mr. Galli, i have tried 3.5x10^4 and 5x10^4 in 24 well plates but the cells are not ready after two days, and they are taking three days to get to required confluency.
