I'm doing a screening on lysates with unknown properties against virus on cell culture. I need to make sure the lysates are sterile so as not to contaminate my cell cultures. What is the best method to sterilize the lysates which potentially contain peptides?
You could try a filtration on 0.2µm filter. For the sterilization, it would work but I do not know if your peptide of interest will not be absorbed into the filter. It exist some low binding filters.
Did you already tested the lysates to see whether they can indeed contaminate? I mean put some lysate in culture media and incubate it mimicking the conditions planed for your experiments.
As mentioned above filtration would be ideal, but if this isn't feasible you might also
Try antibiotics in your culture or preincubation of the lysate with antibiotics before its use on culture.
