We are working with prostate cancer cell lines (PC-3, 22Rv1), and breast cancer cell lines (MCF-7, 4T1) in our lab. Is there an apoptosis inducer that you can suggest to be used as a positive control for these cells?

Hi Jan I tried to induce apoptosis in the following cell lines: Hepatic stellate cells with whom I used TNFalfa+Fas lingand, Cholangiocarcinoma cells with whom I used doxorubicin or arsenic thrioxide. Regards Alessandra
I have read a few papers using different method including 40 nM Taxotere for 0.5 - 24 hours). But, TNFa may be more straightforward and classic if there is FAS-ligand present.
We use puromycin or staurosporine as positive controls for apoptosis. Best concentration will vary with cell type, you would test out a few, but around micromolar levels.
We as Dorothy C Bennett use staurosporine as apoptosis sinducer in MCF-7 and OVCAR-3 cells
I personally have experience with paclitaxel and MCF7 cells. Usually we add 100nM to the culture medium and grow the cells for another 48 hours and check them for apoptosis.
You can use as written previously staurosporine but you can use also C2-Ceramide, doxorubicin, 5-fluorouracil...

Do you have any experience with etoposide as apoptosis inducer?
Narciclasine could also be a good "candidate" for inducing apoptosis in both PC-3 and MCF-7 cancer cells (see the attached article).
