I want to characterize an anti-cancer drug. So, I want to test how extracellular lactate concentrations change due to treatment by my anti-cancer compound. I want to buy a kit but the choice of the kit depends on the extracellular lactate concentrations. Can someone suggest extracellular lactate concentration in cancer cells.
We have measured lactate production in a culture of hybridoma cells (Duval et al, J Immunol methods 134, 1990, 177-185) and found in a large scale culture that lactate concentration in the culture medium may reach up to 10 mMoles/L. Similar experiments carried out in a rat cancer line (PC 12, pheochromocytoma) gave values around 3-5 mMoles/L

I am planning on buying a kit which measures lactate concentrations of about 3000um. Do you think it would be a good idea to do so.
I think so, if theconcentration of lactate in your culture supernatant you may test first a range of dilution 1/3, 1/10, 1/30.... in fresh medium or in a buffer.

Lactate concentration in the extracellular environment depends fundamentally from the type of metabolism of the tumor: fermentative or OXPHOS or both and also from the presence or absence of hypoxia and the expression of lactate transporters like MCT1 and MCT4. So as you may see, the concentration of lactate at the extracellular level depends from the type of cell you are studying and how advanced is the tumor.
If you are interested in the lactate production by tumors, I would recommend to do what Otto Warburg did: use ascitis Ehrlich tumor cells and measure lactate in the ascitic fluid.
