What are the advantages/disadvantages for each in quantifying intracellular protein levels?
Hi, both methods can be used for protein quantification but ELISA is more reliable. This article may be helpful.

Flow cytometry is rather for cell based assays. Also good for the detection of intracellular or membrane proteins. If you need to quantify the level of soluble protein, ELISA is cheaper, easy to learn and more practical.
Preferably assays involving flow cytometry. Thanks.
