Can anyone help: what is best way to prove or show the relationship between Caspase 7 induction and cell signalling pathway or other cellular proteins in PTEN knocked-out colorectal cancer cell lines? I found the caspase 7 induction when I treated PTEN-negative cells with my drug to inhibit one of the DNA repair protein. I checked p21, caspase 3, 2, 9, PARP, LC3 but did not get any expression except capase 7. Therefore, need to dig out further based on caspase 7 based apoptosis. Any help will be highly appreciated. To to include EMT , it will connect her2. As you got caspase 7. But not other . Check parp activity it is also related with apoptosis. Good luck
