Anyonw who knows about Standard/Optimized protocol for clonogenic assay using siRNA?

Colony formation capacity of cells suing siRNA. As cells are incubated for 7 to 14 days, by adding FBS it reduces or revert transfection. Please find attached my optimized protocol.
