I would like to learn the measurement ways for the stemness capacity of the cultured cancer cell. I could not decide what is the best way to do this efficiently. Could anybody recommend the best method of measurement that are tried by you? Thank you.

There is no strict method for measuring stemness, rather it is a combination of several approaches; one of them is to test the expression of specific markers (by qPCR and/or Western) responsible for stem-like qualities for specific cancer type. In parallel, you may compare the capacity of the cells to form tumourspheres in non-adherent conditions. In addition, you may even side-population of cells by FACS. Yet, no universal method is appropriate and better if you try them all.

In cancer cells, studying stemness mostly answers for their subpopulation of cancer stem cells-like cells and their properties.
It is always self-renewal, multilineage potential as well as EMT, Drug resistance, chemo-resistance and many times proliferation, migration/invasion enriched abilities describes about overall stemness of any cancer cells.
Based on the types of cancer, you will get different markers lists which are linked to cancer stemness, like we do work on breast cancer stem cells so check for CD44+, CD24-, ALDH+.
you can prefer Flow cytometry and qPCR to analyse stemness.
