For my thesis work, I will be isolating exosomes from blood. However, the samples will be collected in a remote location and will not be processed immediately. What are the optimal conditions for preservation of exosomal integrity and for how long do they remain stable under these conditions?
If you plan to store your samples for few days you may keep them at 4degree Celsius. However for storage times longer than a week it would be better to freeze the samples, and thaw them on ice just before isolating exosomes. It would be better to avoid repeated freeze thaw cycles.
We resubmit extracted exosomes in PBS at save them in aliquots at -80C. These aliquots can be preserved for months for further cell treatment and/or RNA/DNA/protein extraction.

if you work with blood samples and want to save them for the further exosome isolation:
1) separate plasma or serum;
2) aliquote your plasma/serum samples (0.4-0.5 ml in each aliquote) in cryogenic tubes;
3) save them in a liquid nitrogen cell storage.
You can keep your serum/plasma samples for years with no changes in exosome qualities.
For a few days, we can store at 4oC, but for a longer time we can store at -80oC for a few months and liquid N2 for a longer time. I would suggest thawing the sample on ice. Freezing causes some loss of exosome but still, a good fraction can be obtained.
