It's very easy and you can do it just by spectrophotometric assay.

You can do it by titration or using enzymes. You don't have to use HPLC and MS.
The enzymatic method is typically more sensitive than the titration method.
Glucose 6 Phosphate Dehydrogenase (enzyme) acts on Glucose-6-Phosphate in the presence of NADP to give Gluconate-6-Phosphate and NADPH:
Glucose6P + NADP + (enzyme) --> Gluconate6P + NADPH + (enzyme)
You can measure Abs. of NADPH at wavelength of 340nm using a spectrophotometer. NADPH produced is equimolar i.e. 1M Glucose6P give rise to 1M NADPH.
But you want to look at Glucose1P, which I suppose you could convert using phosphoglucomutase.
Also it depends what you mean by "among other sugar fragments".
