I have been using Ambion MEGAscript RNAi kit for in vitro transcription that yields good results but after the purification process suddenly the dsRNA value plummets significantly downwards. I have trying yo purify by 5M ammonium acetate method and i have good results. In one case 8 micrograms/microlitre.
