I stumbled over an interesting observation:

For some aging-unrelated purpose I overexpressed a gene of interest (GOI) in HEK293 cells (not 293T!). While the untransfected, wildtype cells underwent morphological changes and eventually died at passages x+25 to x+30, the transfected cells kept going to passage 40+ without any problems. (The x stands for the undisclosed number of passages by the vendor before the cells arrived in our lab.)

Currently, I try to repeat the experiment with better controls: a) untransfected wt cells, b) cells stably transfected for empty expression vector and c) cells stably transfected GOI-overexpression vector.

Right now, these cells are at passage x+12.

Any ideas on how to mount an age-accelerating challenge to the cells?

What markers are good to look at? I know that there is SA-beta-Galactosidase. Any further ideas?
