I am working on antigen vaccine delivery system targeting dendritic cell. When I refer some paper to search the dendritic cell (DC)model for in vitro targeting efficacy test of my vaccine delivery system, I realize some papers use DC isolated freshly from femur bone marrow whereas some paper use JAWS II (also one type of bone mature derived dendritic cell). I am confusing which is the difference in purpose of using 2 types of these cell lines. As far as I've known, JAWS II is immortal cell. In my opinion, it should be more convenient to use JAWS II cell line for in vitro experiment. Why do some papers use 2 cell lines for the same purpose which is testing their DC-targeting peptide?
Ex vivo DC is a term to identify freshly extracted DCs from mice / humans.
In vitro derived DCs (mono-DCs / BMDCs) are generated in vitro from a precursor (monocytes or bone marrow) using cytokines.
The JAWS II cell line is neither. These are immature DCs immortalised from p53-deficient B6 mice. Therefore, these cells may harbor inherent differences derived from the p53-deficiency or from the process of immortality.
Using cell lines is often convenient and cheaper, but the findings from the experiments may not reflect events occurring in vivo. Therefore if using a cell line to establish a major result, BMDCs or ex vivo derived DCs are needed to confirm the findings.
In a similar manner, the BMDCs may not reflect the in vivo DC and extracting DCs from spleen or blood may be necessary to confirm the results.

I have worked with JAWS II dendritic cell line in some experiments, This cell line growths slowly in culture and it is dependent of GM-CSF. I guess BMDCs is better than JAWS II, because JAWS has genetic modification that changed its Biology. In my case, we needed higher concentrations of adjuvants to stimulate JAWSII comparing with spleen DCs, Just high quantities of LPS increased CD86, CD80 and MHCII expression in JAWS II.
Actually, in my opinion the best type of DC is DC isolated from spleen by a MACS sorter.

I recently started working with JAWSII cells and I noticed that online primary BMDCs these cells don't protrude dentists or attach after LPS treatment. I treated them with 100ng/ml and 1000ng/ml and saw same results. Can you please let me know if you see the same or if there is better way to mature them?
