Does anyone knows some article that validates that the morphology of vaccinia or pox virus is not affected by UV treatment. Preferably with protocol and EM photos conforming it. I would like to have a protocol to produce a non replicating vaccinia virus with the same structure and morphology as the wild type, using UV as a inactivation method, that only affects the DNA.
I want to have a "dead" virus to follow its migration trough the lymphatic system.

From what I know, the replication can be halted but the infective characteristics stay unaltered. I think you may have to mention more information on why you want to produce non replicating vaccinia. Is it for vector related research or drug delivery ? Probably the research gate community can help you better if that is mentioned. Hope this helps
You can purchase those UV-inactivated VACV from some commercial companies (eg. Advanced Biotechnologies Inc.). The virus is morphologically intact (as verified by TEM) and it is not infectious. I believe you will have to do some legal paperwork before buying it.
