I would like to know whether I can use RT-PCR to check mRNA level of possible downstream genes in the crispr mutants of Arabidopsis. The mutation type of the genes in these crispr mutants are small indels (1 ~ 10bp) that result in pre-mature stop codons of their protein sequence.
I learned that unlike T-DNA mutants in Arabidopsis, which most time have disruption in transcription, the transcription level of crispr mutants with indels, however, may not be affected.
Are the mRNA level of downstream or upstream genes in crispr mutants also not affected unless there is a big deletion in the mutants??
