In order to use the NEB Tm calculator, is it the addition of the forward and reverse primer concentration that is required or just one of them?
you should estimate each primer separately
first calculate for each primer individually, then calculate together. Also correctly enter the polymerase from the drop down menu and primer concentration to get most probable Ta.
Alternatively you can also use the IDT DNA Oligoanalyzer tool.
