I dissolved the formulation in DMSO, I get a clear solution so I started with the assays but I could see certain abrupt fluctuations instead of linearity in the results. This made me suspect the possiblity of protein interference.
If yes, I would be delighted to get suggestions for which method of deproteination could be effective with DMSO as a solvent.
are the herbal formulation contains protein? If not, then its not need to go for deproteination. Fluctuation in assay results may also be due to degradation of herbal constituents in DMSO Solvents.
Please keep in mind that DMSO itself exerts also some antioxidant activity and tehrefore might interfere with the assay. Moste studies nowadays suggest to avoid DMSO...instead two assays are suggested: One for the lipophilic antixodants in extracts made in lipophile solvents and one done from aqueous extracts...
