I would like to compare the effect of 4 different TKIs on 20 different cells lines. Should I adjust the seeding cell number for each cell line (based on its growth curve) or should I start with the same number of cells for each cell line i.e. 500 cells for each?

The question is are you going to treat the cells at confluence or at growth phase. Some of the inhibitors do not show efficacy at confluence. What is your scoring method?
You may want to reach a semi-confluence state in a fixed time. To achieve that you may to adjust the seeding density

if your number of different cell lines is 20 - you will almost for sure have to seed different numbers - as you wrote depending from the growth curve (or doubling time). Alternatively it is also possible to seed equal numbers and start the drug testing to different time points depending on the targeted % confluency. But this is not prefered by most people since it is not as practical as doing 5, 10 or even 20 cell lines at once.
For TKIs it is suitable to use cells in the "close-to-log"-phase. As Ravi wrote, semi-confluent will do the job.
