has anyone any general thoughts on the effects of agar firmness on plant development using apical stems in firm versus soft agar? Since auxins produced in the meristems and can be transported down the stem , if a cut off stem is placed in the agar then is it possible that in a firmer agar that the endogenous auxins can get sequestered around the cut end where its sitting in the agar since they are less able to diffuse thru the medium. changing auxin/cytokinin ratios and or concentrations with those already added to the medium. I'm tissue culturing Nepenthes and other carnivorous plants. thanks steve
