Hi, I am looking for suitable cell line model for my new experiment and THP-1 is one of my choice to test. If someone has these cells cultured under ATCC-recommended condition (without additional glucose, FBS or extra supplements) and is willing to share I will appreciate it a lot. The Czech Republic lab is advantageous but not restricted (I do not know how frequent this cell line is and how many people work with it). Thank you all in advance for any feedback. P.

Our lab recently started working with these cells. You must be aware that these are undifferentiated cells and grow in suspension. These cells need to be grown at low density as their overcrowding will lead to cluster formation which is not for their phenotype. According to the special instructions from ATCC , these cells should not be subcultured 2 in the month and should not be used after passage 8. Some labs only use them till passage 5, but the information that I am giving is what I got directly emailing ATCC. I'm attaching a pdf from Dr. Bowdish's lab that I found helpful as it's very detailed and will help you get started with your work . You will encounter the need to change the protocol as you go ahead with your work . Also, the protocol which I'm sharing does not include information about antibiotics. I have used 0.25x pen-strep antibiotics in my culture media to avoid contamination and the freezer stocks don't need extra FBS , it can be stored in complete media and 5%DMSO.
As these are suspension cells , it will be difficult to judge their number , just be looking at them, so it will be better to homogenise the suspension and count the cells by Trypan blue dye exclusion method to know the exact number of cells, Don't let them grow over 1 million/ ml as they start clumping which is not good at all.
