We have made a PMMA cartridge (containing channels and reaction chamber) and we have sealed it completely with Ethanol 96% in 90℃.

In order to coat the surface of reaction chamber with antibody I do following steps:

1) washing channel with EtOH

2) treatment of channel with NaOH 1M 1 hour at 55℃.

3) washing with DI water

4) treatment with Polyethyleimine (PEI) and glutaraldehyde (GA) pH 7

5) Antibody coating with a carbonate/bicarbonate buffer pH 9.5 at 4℃ overnight

I faced with some problem:

1) the bonding of PMMA layer was weaken after washing and coating steps.

2) cracking of PMMA after washing and coating steps.

3) trapping of fluid in angles of rectangular channels and reaction chamber.

I will be appreciated to help me in these issues.
