Hello, recently I am having some difficulties with maintaining the cell culture of primary cells isolated from a skin biopsy (normal and malignant). The coating of culture plates with Matrigel (a mixture of basement membrane proteins) doesn't seem to be sufficient.
Next on my list to try is coating plates with collagen I and investing in special plates for primary cultures (like Primaria from Corning).
Maybe some of you have some hands-on experience and are willing to offer a piece of advice? I would be really grateful!
Hello Ewelina, have you tried fibronectin or collagen instead? For an experiment I had to expand human skin fibroblasts and collagen worked very well (even for senescent cells). Also, have you considered growing your cells on a cell insert? -(cells grow better and you can perform air-liquid interface cultures)- When using inserts, pore size is a critical factor. Thank you for your suggestions. Collagen coating was my next idea to try so it's nice to get a confirmation that it may work.
In the case of inserts - we are trying this too but in a different context - we're studying the influence of cancer cells on normal cells... But it could serve a double purpose - help cancer cells and give us answers about their interaction.
